Processing proteases of viral glycoproteins
Description of scientific projects:
Proteolytic Activation of Viral Glycoproteins
Subcellular transport and maturation of the subtilisin - like
endoprotease furin
Enveloped viruses, such as influenza, and HIV, posses glycoprotein spikes on their surface which mediate binding to cellular receptors and fusion of viral envelope with cellular membranes. To gain fusion capacity, the majority of viral glycoproteins is post-translationally cleaved by cellular proteases. Among them, the eukaryotic, subtilisin-like furin is the key enzyme for cleavage at multibasic cleavage sites. We found that furin, an integral membrane protein alike viral glycoproteins, is transported along the exocytotic pathway where it accumulates at the trans-Golgi-network (TGN). We are interested in TGN-localization signals and signal recognition proteins.
